Extended loci histocompatibility matching in HSCT—Going beyond classical HLA

نویسندگان

چکیده

Unrelated haematopoietic stem cell transplantation (HSCT) has evolved from an experimental protocol to a potentially curative first-line treatment in variety of haematologic malignancies. The continuous refinement protocols and supportive care paired with ongoing achievements the technological field histocompatibility testing enabled this transformation. Without doubt, HLA matching is still foremost criterion for donor selection unrelated HSCT. However, HSCT-related complications occur frequently, often resulting patients suffering severely or even dying as consequence such complications. Current literature indicates that other immune system modulating factors may play role setting In review, we discuss current clinical evidence possible influence nonclassical antigens HLA-E, HLA-F, HLA-G well HLA-like molecules MICA MICB,

Major Histocompatibility Complex and Hematopoietic Stem Cell Transplantation: Beyond the Classical HLA Polymorphism

Allogeneic hematopoietic stem cell transplantation (HSCT) represents a curative treatment for many patients with hematological malignant or non-malignant disorders. Evaluation of potential donors for HSCT includes a rigorous assessment of the human leukocyte antigens (HLA) match status of family members, and the identification of suitable unrelated donors. Genes encoding transplantation antigen...

HLAMatchmaker: a molecularly based algorithm for histocompatibility determination. V. Eplet matching for HLA-DR, HLA-DQ, and HLA-DP.

This report describes the design of the eplet version of HLAMatchmaker to determine class II compatibility at the structural level. This matching algorithm is based on the hypothesis, developed from molecular modeling of crystallized antigen-antibody complexes, that functional epitopes are represented by patches of surface-exposed nonself-amino acid residues surrounded by residues within a 3-A ...

HLA matching

INTRODUCTION — The evidence that antigens of the HLA system provide the major barrier to acceptance of renal transplants was first obtained with living-related donor transplants. Graft survival was superior in sibling pairs having both the same serologically defined HLA antigens and a nonreactive in vitro mixed lymphocyte proliferative response when compared to randomly matched cadaveric donors...

Systemic lupus erythematosus and the extended major histocompatibility complex--evidence for several predisposing loci.

OBJECTIVE Systemic lupus erythematosus (SLE) is an autoimmune disease reported to be associated with several alleles in the HLA complex. The purpose of this study was to systematically examine the extended HLA complex (xMHC) in order to get an overview of the primary predisposing genetic factors. MATERIALS AND METHODS One hundred and sixty-four SLE patients and 254 healthy, unrelated controls...

Dominant Sequences of Human Major Histocompatibility Complex Conserved Extended Haplotypes from HLA-DQA2 to DAXX

We resequenced and phased 27 kb of DNA within 580 kb of the MHC class II region in 158 population chromosomes, most of which were conserved extended haplotypes (CEHs) of European descent or contained their centromeric fragments. We determined the single nucleotide polymorphism and deletion-insertion polymorphism alleles of the dominant sequences from HLA-DQA2 to DAXX for these CEHs. Nine of 13 ...
